Influence of Splenic Operation on Bacterial Sepsis in Rats




Abstract
Male Sprague-Dawley rats weighing between 200-250g were allocated two sections-bacterial clearance or phagocytic activity with technitium 99m-tin colloid(TcTC) uptake. Each section was performed following procedures : total splenectomy(TS).
hemisplenectomy(HS), splenic artery ligation(AL), intraperitoneal splenic autotransplantation(AT), or sham laparotomy(LA). At 3, weeks after the operation, 107 Staphylococcus aureus. 10 Streptococcus pneumoniae, 10 E. coli, 106 Bacteroids
fragilis,
or
10 Klebsiella pneumoniae were injected intraperitonealy into the first section of the rats. Blood samples from femoral veins for quantitative culture were obtained at serial after bacterial challenge. TS group had difficulties in all bacterial
clearance
compared with other groups. HS and LA group developed early bacteremia and cleared all bacteria from the blood stream soon but Bacteroids. AL group demonstrated reduced clearance for Staphylococcus and AT caused a decreased in clearance for all
bacteria
except Klebsiella Splenic phagocytic function was determined by measuring radiocolloid uptake 15 minutes after TcTC injection. Splenic phagocytic function[(cpm spleen/cam liver) 100] in HS. AL and AT groups significantly decreased compared with
LA
group(p<0.01). Phagocytic index expressed. as (cpm spleen/g) (cpm liver/g), was significantly higher in HS group than in LA group, and AT resulted in significantly lower phagocytic index than LA did. Light microscopic examination demonstrated
structural
changes of spleen from rats with splenic artery ligation and autotransplantation. We conclude that partial splenectomy with preserving splenic artery neither affect bacterial clearance, nor phagocytic activity, however, splenic artery ligation
caused a
delayed bacterial clearance, but maintained phagocytic activity. Bacterial clearance and phagocytic activity remains reduced after splenic autotransplantation.
